چکیده

Baicalein, one of the major flavonoids in Scutellaria baicalensis (Chinese Skullcap), is well known for its effects on cell proliferation, apoptosis, and inflammation. Here we show that baicalein also inhibits the adipogenesis of 3T3-L1 preadipocytes. Baicalein inhibited triglyceride accumulation during adipogenesis and significantly decreased the mRNA expression of fatty acid-binding protein (FABP), a marker of adipogenesis. Microarray analysis revealed that several genes, which are differentially expressed during adipogenesis, were modulated by baicalein treatment in 3T-L1 cells. The expression of FABP, apolipoprotein D, and insulin-like growth factor 2, which was markedly up-regulated during adipogenesis, was down-regulated by baicalein. Cyclooxygenase (COX)-2 mRNA expression, which was decreased during adipogenesis, was up-regulated by baicalein. These COX-2 mRNA expression patterns were mirrored by the expression of COX-2 protein and its enzymatic activity. NS-398, a COX-2 inhibitor, partially abrogated the baicalein-induced inhibition of adipogenensis. Thus, the anti-adipogenic effect of baicalein may be mediated by its ability to enhance the expression of COX-2, which is normally down-regulated during adipogenesis.

Arachidonic acid-dependent inhibition of adipocyte differentiation requires PKA activity and is associated with sustained expression of cyclooxygenases.

Arachidonic acid inhibits adipocyte differentiation of 3T3-L1 cells via a prostaglandin synthesis-dependent pathway. Here we show that this inhibition requires the presence of a cAMP-elevating agent during the first two days of treatment. Suppression of protein kinase A activity by H-89 restored differentiation in the presence of arachidonic acid. Arachidonic acid treatment led to a prolonged a...

Adipocyte differentiation of 3T3-L1 preadipocytes is dependent on lipoxygenase activity during the initial stages of the differentiation process.

Adipocytes play a central role in whole-body energy homoeostasis. Complex regulatory transcriptional networks control adipogensis, with ligand-dependent activation of PPARgamma (peroxisome proliferator-activated receptor gamma) being a decisive factor. Yet the identity of endogenous ligands promoting adipocyte differentiation has not been established. Here we present a critical evaluation of th...

Suppression of Very Early Stage Of Adipogenesis by Baicalein, a Plant-Derived Flavonoid through Reduced Akt-C/EBPα-GLUT4 Signaling-Mediated Glucose Uptake in 3T3-L1 Adipocytes

Baicalein has been used as a Chinese medicine, and is an abundant plant flavonoid present in fruits and vegetables. Here, we examined the effects of baicalein in adipogenesis and investigated its molecular mechanism in adipocytes. Baicalein lowered the intracellular lipid accumulation and decreased the transcription levels of the adipocyte-specific genes in mouse 3T3-L1 adipocytes. Glucose upta...

Baicalein inhibits the migration and invasion of colorectal cancer cells via suppression of the AKT signaling pathway.

The anticancer effect of baicalein has been known for a number of years. However, its anti-metastatic effect and associated mechanisms in colorectal cancer (CRC) remain unclear. The present study investigated the hypothesis that treatment with baicalein may inhibit the proliferation, motility and invasion of human CRC cell lines via regulation of the protein kinase B (AKT) signaling pathway. Ba...

Inhibition of cancer cell proliferation and prostaglandin E2 synthesis by Scutellaria baicalensis.

Scutellaria baicalensis is a widely used Chinese herbal medicine that has been used historically in anti-inflammatory and anticancer therapy. The purpose of this study is to verify its anticancer activity on head and neck squamous cell carcinoma (HNSCC) in vitro and in vivo and to investigate its effect on cyclooxygenase-2 (COX-2), which converts arachidonic acid to prostaglandin E(2) (PGE(2)) ...
